I think that at that time knowledge of sealing was limited but not non existent as the sand Village had captured shukaku with the help of hashirama or konoha in the armistice treaty they had asked for land for cultivation from konoha and we know that the uzumaki clan is the descendent of Ashura so ....I guess sealing existed before hashirama
